This host strain is a derivative of the Sparrowhawk strain and can be used for bioengineering
where purified products with reduced endotoxicity are desirable. Enterobacterial common
antigen is no longer produced and a UDP N-acetylglucose epimerase has been chromosomally
integrated increasing the availability of UDP-GalNAc. The O-antigen ligase has also been
removed so that recombinant glycans are not removed from Und-P and attached to surface lipid.
The chain length regulators from Streptococcus pneumoniae capsule have replaced the O-antigen
chain length regulator of E. coli, in order to increase polymerization of plasmid expressed
S. pneumoniae capsule glycans. The Campylobacter jejuni pglB oligosaccharyltransferase with a 0.1 strength Anderson promoter
is integrated into the chromosome of the Sparrowhawk strain, between atpI and gidB using CRISPR
technology, for bioconjugation.
